I use the Slink-e on a 100 foot run of CAT-5 cable every day at 38.4k. The length you are trying to acheive is certainly workable. There may be a reliability problem with having so many extensions plugged together. I have had SlinkeServ not a find a Slink-e on rare occasions, but this is typically due to some other piece of software using the COM port. A few months ago I made some changes to the code which does the auto-baud / auto-detection of Slink-es to reduce the time this takes. This may have compromised the reliability of the detection a bit, but it didn't seem to show up here. What % of the time do you have this problem? When it starts happening, does it continue to happen over multiple retries?
